IGF-1 LR3 (Long Arginine 3-IGF-1) is an 83-amino-acid analogue of human insulin-like growth factor 1, carrying an arginine substitution at position 3 and a 13-residue N-terminal extension. These modifications sharply reduce its affinity for IGF-binding proteins while preserving receptor activity, giving the analogue markedly higher biological potency than native IGF-1 in experimental systems.

RESEARCH BACKGROUND
IGF-1 LR3 emerged from work in Adelaide in the early 1990s, where recombinant IGF-1 analogues were engineered to escape the binding proteins that normally sequester IGF-1. The landmark 1992 study showed that LR3-IGF-1 binds IGF-binding proteins poorly while retaining receptor activity — making it substantially more potent than native IGF-1 in cell-based assays.
Since then, IGF-1 LR3 has become a workhorse of growth-factor research and biomanufacturing, most prominently as a media supplement sustaining growth and productivity of CHO and other production cell lines. It remains among the most used IGF-1 analogues in the published literature.
